  • Ritgerð þessi er sett upp í tveimur hlutum:
    Leymus Hochstetter (melgresi) og Triticum L. (hveiti) eru tegundir ættbálksins Triticeae. Melgresi er þekkt fyrir að lifa í umhverfi sem aðrar plöntur þola ekki. Lítil úrkoma, mikið sandfok og selta eru skilyrði sem melgresi þolir vel. Þessir þolnir eiginleikar eru einkar eftirsóknarverðir í landgræðslu og því hefur samþætting þessara eiginleika við hina hefðbundu hveitiplöntu verið í brennidepli. Margar útfærslur hafa verið gerðar á blendingum hveitis og melgresis en í þessari rannsókn er blendingur melhveitis rannsökuð. Með aðferðinni Genomic in situ hybridization (GISH) voru genamengi melgresis og hveitis færð á þreifara og þáttatengd við rannsakaða melhveitið. Út fékkst heillitun 28 hveitilitninga og 14 melgresislitninga. Loks voru 18S-25S ríbósómgen kortlögð með Fluorescence intensity hybridization (FISH) aðferðinni sem sýndi genin Nor-B1, Nor-B2 ásamt ríbósómgen melgresis.
    Saxifraga L. (steinbrjótur) er meðal ættkvísla tvíkímblöðunga sem hafa tekist að taka yfir sig breitt svið útlitseinkenna, breytileika í erfðum, vistfræðilega útbreiðslu og fjölbreytileika á lífsháttum. Í þessari rannsókn voru litningar fjögurra steinbrjótstegunda einangraðir með protoplast-dropping aðferð. Litningarnir voru taldir í flúrsmásjá í DAPI lit. Unnið var með Saxifraga cernua 2n=>30, S.granulata 2n=>30, S. nivalis 2n=60 og S. rosacea 2n=>30. Niðurstöður sýndu fjölbreytileika milli tegunda. Þessi breytileiki mætti útskýra útlitsbreytileika milli tegunda. Samkvæmt heimildum er grunntala Saxifraga L. x=6,7,8,9,10,11 og 13 og er hann því fjöllitna. Þetta gerir plöntuna mjög áhugaverða til rannsóknar, hún vex villt og æxlast eðlilega. Þetta sýnir fram á eðlilega meiósu þrátt fyrir margvíslega fjöllitnun, upplýsingar um genamengi Saxifraga L. er því mjög mikilvæg.

    This thesis is divided into two parts.
    Leymus Hochstetter (lymegrass) and Triticum L. (wheat) are species of the tribe Triticeae. Lymegrass is particularly known for survival in habitats that other plants have difficulty surviving in. Drought, erosion and salinity are all conditions that lymegrass can thrive in. These resistant traits are very much sought after in agriculture, therefore the integration of these traits into the traditional wheat has been accomplished. Many versions of wheat x lymegrass hybrids have been made, the version used in this thesis is wheat x lymegrass hybrid which has been back-crossed with wheat multiple times. With the technique Genomic in situ hybridization (GISH), the genome of this particular plant was labelled with probes from wheat and lymegrass genomes. This resulted in a fully colored green (lymegrass) vs. red (wheat) coloring of its genome. The chromosomes of the plant were counted per genome and together as a whole, the results showed 14 lymegrass and 28 wheat chromosomes. The ribosomal genes 18S-25S were then mapped using Fluorescence in situ hybridization (FISH) approach which resulted in the labelling of the Nor genes Nor-B1 and Nor-B2 as well as the ribosomal genes of lymegrass.
    Saxifraga L. (saxifrage) are one of the genera of dicots that has managed to incorporate a broad spectrum of morphological, genetic, ecological and life cycle differences. In the following research, the chromosomes of four different saxifrage species were isolated by using shoot tip protoplast dropping method and counted using a Fluorescence microscope with DAPI dye. The researched species were Saxifraga cernua 2n=>30, S.granulata 2n=>30, S. nivalis 2n=60 og S. rosacea 2n=>30. Results showed a variable between species. According to other sources the base number Saxifraga L. are x=6, 7, 8, 9, 10, 11, 13 and it is therefore polyploid. Species of Saxifraga L. grow wild and are fertile despite various forms of polyploidy, which makes the genome of Saxifraga L. an important for further studies in plant-genome research.
