    Pontin and Reptin are highly conserved proteins belonging to the AAA+ family of ATPases. They are associated with several multiprotein complexes involved in various cellular mechanisms. In this project, the subcellular localization of Pontin and Reptin, as well as their co-localization with Rpt1, a component of the base subunit of the proteasomal 19S RP, were examined using Drosophila melanogaster as a model. Fluorescently tagged expression clones of Pontin, Reptin, Rpt1 and Rpt6 were constructed and used for the establishment of transgenic fly lines. These fly stocks were used for the controlled overexpression of the proteins in the fly brain using the Gal4/UAS system. Both Pontin and Reptin were found to localize to the cell periphery. Strong distribution of Reptin to the nucleus was also evident. Co-expression of Pontin and Reptin influenced the distribution of Reptin. Mutations introduced into the conserved Walker domains of Pontin and Reptin led to alterations in cell shape in the Reptin mutant when co-expressed with Pontin. Strong co-localization of Pontin and Reptin with Rpt1 was evident in several regions. Interestingly, all proteins showed localization to mitotic spindles and the microtubule cytoskeleton. Knock-down of Reptin, Rpt1 and Rpt6 led to abnormal photoreceptor projection patterns. Future experiments focusing on the interaction of Pontin, Reptin and 19S RP subunits, their connection to the microtubule cytoskeleton, as well as their role in nervous system development will therefore be of interest.
