  • Skaði á hornhimnu vegna slysa og sjúkdóma er ein algengasta orsök blindu og algengasta líffæraígræðslan. Skortur á hornhimnum frá látnum gjöfum kallar á nýjar leiðir til að lagfæra hornhimnur. Hér var leitað leiða til að endurbæta hornhimnur með stofnfrumum úr mennskum fósturvísum. Markmið þessarar rannsóknar var að skoða hvort að stofnfrumur úr fósturvísum tjáðu ensímið Na+/K+-ATPase eftir ræktun á Descemet himnu. Stofnfrumur af stofnfrumulínu SA181 úr mennskum fósturvísum voru ræktaðar og settar á Descemet himnuna sem hafði verið hreinsuð að hluta af endothelium frumum. Frumurnar voru ræktaðar í allt að 21 dag á Descemet himnunni.Stofnfrumunum hafði verið breytt erfðafræðilega til að tjá flúrljómandi grænt prótein og voru því auðþekkjanlegar í gegnum ræktunarferlið. Til að greina hvort stofnfrumurnar breyttust í endothelium frumur var monoclonal mótefni fyrir Na+/K+-ATPase notað. Myndgreining var gerð með Nikon flúrljóm smásjá með DAPI, TRITC og FITC filterum. Niðurstöður ónæmisvefjalitunar voru að ágræddu frumurnar formuðu endothelium líkt frumulag og tjáðu ensímið Na+/K+-ATPase. Niðurstöðurnar sýna að mögulegt er að græða stofnfrumur úr fósturvísum á Descamet himnu þegar endothelium frumur hafa verið fjarlægðar að hluta, og hægt er að fá þessar frumur til að sérhæfast í endothelium líkar frumur in vitro.

    Corneal damage due to disease and accidents is one of the leading causes of blindness in the world, and one of the most frequently transplanted organs. Corneas are procured from deceased donors and are therefore in extremely limited supply. This lack of corneas calls for new ways of healing corneas and in this research human embryonic stem cells were used to regenerate the Descemet membrane of the cornea. The goal of this study was to assess whether stem cells expressed the enzyme Na+/K+-ATPase after being grown on the Descemet membrane. Stem cells of stem cell line SA181 which had been genetically transformed to express green fluorescent protein were easily recognizable throughout the transfection procedure. The stem cells were cultured and put on the Descemet membrane that had been partially scraped clean of endothelial cells. To determine weather the stem cells changed into corneal endothelial cells, a monoclonal antibody for the enzyme Na+/K+-ATPase was used. The expression of Na+/K+-ATPase, was analysed with immunohistochemistry and photographed with a Nikon fluorescence microscope equipped with DAPI, TRITC and FITC filters. The results show that the transplanted cells formed an endothelial- like cell layer and expressed Na+/K+-ATPase. The results show that it is possible to transplant human embryonic stem cells onto a Descemet membrane and get the cells to differentiate into endothelial like cells in vitro.
