  • Inngangur: Náttúrulegar drápsfrumur (NK frumur) eru hluti af náttúrulega ónæmiskerfinu og gegna hlutverki í fyrstu vörnum gegn sýkingum. Sýnt hefur verið fram á að með því að auka magn ómega-3 fitusýra í fæði manna geti það haft áhrif á bólguhjöðnunarferlið og þar með komið í veg fyrir myndunar á krónískri bólgu. Fyrri rannsóknir á vakamiðlaðri bólgu hafa leitt í ljós að mýs sem fá fiskolíuríkt fæði hafa aukið magn NK frumna snemma í bólguferlinu og einnig hraðari bólguhjöðnun. Áhrif fitusýra á NK frumur í mönnum hafa lítið verið rannsakaðar m.t.t. mögulegra áhrifa þeirra á bólguhjöðnunarferlið.
    Markmið: Markmið verkefnisins var að kanna áhrif ómega-3 og ómega-6 fitusýra á tjáningu NK frumna í mönnum á ýmsum viðtökum sem m.a. taka þátt í bólgumyndun og bólguhjöðnun.
    Aðferðir: NK frumur voru einangraðar in vitro úr einkjarna frumum og ræktaðar í návist arakídónsýru (AA), eikósapentaensýru (EPA) og dókósahexenónsýru (DHA) eða viðmiðs (DMSO) og síðan ræstar með IL-12 og IL-15. Áhrif fitusýranna á tjáningu ýmissa NK frumu viðtaka var metin með frumuflæðisjá.
    Niðurstöður: Fitusýran EPA minnkaði hlutfall NK frumna sem tjáðu NKG2D samanborið við viðmið en hafði engin áhrif á tjáningu annara NK frumu viðtaka. Ræktun NK frumna með fitusýrunni DHA jók meðalflúrljómun frumna sem tjáðu viðtakana CCR7 og CD62L, en EPA jók einnig CCR7 meðalflúrljómun frumnanna.
    Ályktanir: Þar sem ómega-3 fitusýrurnar hækkuðu ekki tjáningu á NKG2D eða NKp46 viðtökunum, sem báðir hafa verið tengdir við aukinn sjálfvirkan frumudauða, verður að álykta að þessir viðtakar hafi að öllum líkindum ekki áhrif á aukningu á bólguhjöðnun sem sást hjá músum sem voru fóðraðar með fiskolíuríku fæði. Þá bendir aukin tjáning á CCR7 og CD62L hjá NK frumur ræktuðum í návist ómega-3 fitusýranna EPA og DHA til þess að þær NK frumur séu líklegri til að ferðast til eitla þar sem þær geta hugsanlega komið óbeint að bólguhjöðnunarferlinu í gegnum Th1 frumur.

    Introduction: Natural killer (NK) cells are part of the innate immune system and are known to act as the first line of defence during an infection. It has been shown that human diet, rich in omega-3 fatty acids, can affect resolution of inflammation and thereby prevent the formation of chronic inflammation. Previous studies on antigen-induced inflammation have shown that mice receiving dietary fish oil have increased levels of NK cells early in the inflammatory process and more rapid resolution of inflammation. Effects of fatty acids on NK cells in humans with regard to their potential role in the process of resolution of inflammation has not been studied.
    Aim: The aim of the study was to investigate the effect of omega-3 and omega-6 fatty acids on expression of various receptors, e.g. involved in induction and resolution of inflammation, in human NK cells.
    Methods: Human NK cells were isolated from peripheral blood mononuclear cells and cultured in the presence of arachidonic acid (AA), eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) or control (DMSO) and then stimulated with IL-12 and IL-15. The effect of fatty acids were assessed by measuring the expression of surface receptors by flow cytometry.
    Results: NK cells cultured in the presence of EPA had decreased NKG2D expression compared with NK cells cultured with DMSO, but the fatty acids had no other effects on the expression of other NK cell receptors. Culturing NK cells with the fatty acids EPA and DHA increased expression levels of the chemokine receptor CCR7, and culturing NK cells with DHA increased also the expression levels of the homing receptor CD62L.
    Conclusion: As the omega-3 fatty acids did not cause an increase in the expression of the apoptosis-associated receptors NKp46 and NKG2D on NK cells, it may be concluded that the receptors are unlikely to be involved in the increase in resolution of inflammation observed in mice fed on fish oil diet. The results suggest that NK cells that have increased expression of CCR7 and CD62L when cultured in presence of omega-3 fatty acids EPA and DHA are more likely travel to the lymph nodes where they may have indirect effects on the inflammatory process through Th1 cells.
