    Autophagy is a degradation process required for cellular homeostasis. The process is a means for the cell to retrieve energy and nutrients under stress conditions. Autophagy is involved in a variety of cell related processes and diseases, from cancer to development and aging. Three distinct autophagy pathways are known: macroautophagy, microautophagy and chaperone-mediated autophagy. Of the three pathways, macroautophagy is the most studied pathway and thought to be the most prevalent one. During macroautophagy, a double membrane, termed phagophore, forms and expands until it encloses to form a sealed vesicle called an autophagosome. Finally, the autophagosome fuses with a lysosome where the cargo is degraded before its basic components are released into the cytosol.
    Autophagy-related protein 7 (ATG7) is a key protein in the elongation of the phagophore membrane. The protein facilitates the lipidation of LC3/GABARAP which is essential for the phagophore expansion and the morphological transition of the phagophores membrane sheet to the autophagosomes’ sealed vesicle. The lab recently identified an isoform of ATG7 that is not able to lipidate LC3/GABARAP. This isoform is termed ATG7(2), opposed to the canonical ATG7(1). The aim of this project was to a) analyze the expression of the ATG7 isoforms in human tissue and b) characterize possible functions of ATG7(2).
    Analysis of ATG7 mRNA expression data revealed an increased expression of ATG7 in tumors compared to normal tissue. This increased expression of ATG7 was driven by an increase in ATG7(2) expression, rather than ATG7(1) expression, especially in metastatic tumors. This implicates a role of ATG7(2) in tumors and metastasis. This is in accordance with the groups functional studies which have revealed a possible role of ATG7(2) in migration.
    Blood was the only normal tissue where ATG7(2) expression was higher than ATG7(1) expression. According to ATG7 mRNA expression data of individual hematopoietic cell types, the highest expression of ATG7 is in monocytes and neutrophils. Although the values for the mRNA expression in hematopoietic cells are of total ATG7 expressed, and not of individual transcripts, enrichment analysis of ATG7(2)’s protein interactome further supported the hypothesis that ATG7(2) is most likely to be expressed in monocytes and/or neutrophils. Human peripheral blood mononuclear cells (hPBMCs) from healthy individuals were analyzed for ATG7(2) expression. RT-qPCR showed that mRNA of both isoforms is expressed in the cells. Flow cytometry showed that ATG7 is expressed as protein in hPBMCs, most likely monocytes. Immunoprecipitation of ATG7 from isolated monocytes, followed by western blot, confirmed its presence in monocytes.
    We have characterized the expression of an ATG7 isoform, that does not have a known autophagy function, in blood and tumors. ATG7 is known to have autophagy-independent function, related to apoptosis and phagocytosis. It will be interesting to further characterize the role of ATG7(2) and whether the autophagy-independent roles of ATG7 might be through ATG7(2). The characterization of ATG7(2) will also make it a useful tool to investigate ATG7-independent macroautophagy, which has been described but is not well known. ATG7 is an important autophagy protein and an essential protein for mammals and the characterization of its isoforms will provide us with fundamental knowledge about the autophagy process and its connection to other cellular pathways.

  • Sjálfsát er niðurbrotsferli í frumum og er mikilvægt fyrir almennt heilbrigði þeirra. Ferlið er leið frumunnar til að endurheimta orku og næringu við streituaðstæður. Sjálfsát tengist ýmsum ferlum og sjúkdómum, allt frá krabbameinum til þroskunar og öldrunar. Þrír sjálfsátsferlar eru þekktir: stórsjálfsát, smásjálfsát og vörslupróteins-sjálfsát. Af þessum ferlum er stórsjálfsát þekktast og talið vera algengast. Við stórsjálfsát myndast tvöföld himna sem stækkar þar til hún verpist og myndar lokaða bólu, sjálfsátsbólu. Að lokum rennur sjálfsátsbólan saman við leysikorn þar sem farmurinn er brotinn niður í grunnbyggingareiningar sem síðan er seytt út í frymisvökvann.
    Autophagy-related protein 7 (ATG7) gegnir lykilhlutverki við myndun sjálfsátsbólunnar. Próteinið greiðir fyrir bindingu lípíðs við LC3/GABARAP prótein sem er nauðsynlegt fyrir lengingu himnunnar og verpingu hennar til að mynda sjálfsátsbóluna. Hópurinn greindi nýlega frá ATG7 ísóformi sem getur ekki tengt lípíð við LC3/GABARAP. Þetta ísóform er nefnt ATG7(2) en það ísóform sem hefur sjálfsátsvirkni er nefnt ATG7(1). Markmið þessa verkefnis var að a) greina tjáningu ATG7 ísóformanna í mannavefjum og b) greina mögulega virkni ATG7(2).
    Greining á ATG7 mRNA tjáningu sýndi fram á aukna tjáningu á ATG7 í æxlisvefjum, miðað við heilbrigða vefi. Þessa auknu tjáningu má rekja til aukinnar tjáningar á ATG7(2) fremur en ATG7(1), sérstaklega í meinvörpum. Þetta bendir til þess að ATG7(2) gegni hlutverki í æxlum og meinvarpsmyndun. Það samræmist tilraunum hópsins sem hafa sýnt fram á mögulegt hlutverk ATG7(2) við frumuflakk.
    Blóð var eini heilbrigði vefurinn þar sem tjáning ATG7(2) var hærri en tjáning ATG7(1). Samkvæmt gögnum um mRNA tjáningu ATG7 í einstökum gerðum blóðfrumna, er hæsta tjáning ATG7 í einkjörnungum og daufkyrningum. Þó gögnin um mRNA tjáningu í blóðfrumum nái yfir heildartjáningu á ATG7, fremur en einstök umrit, styður greining á ATG7(2) prótein-prótein víxlverkun þá tilgátu að tjáningu ATG7(2) sé líklegast að finna í einkjörnungum og/eða daufkyrningum. ATG7(2) tjáning var greind í sýnum af hnattkjarna frumum úr blóði (hPBMCs) heilbrigðra einstaklinga. RT-qPCR sýndi að frumurnar tjáðu mRNA af báðum ísóformunum. Greining með flæðifrumusjá sýndi fram á tjáningu ATG7 próteins í hnattkjarna frumum, líklegast einkjörnungum. Mótefnaúrfelling á ATG7 úr einangruðum einkjörnungum staðfesti tjáningu einkjörnunga á ATG7 próteini.
    Við höfum greint tjáningu á ísóformi ATG7 sem hefur ekki þekkta sjálfsátsvirkni, í blóði og auk þess í æxlum. Áður hafa verið greind hlutverk ATG7 sem eru óháð sjálfsáti en tengjast stýrðum frumudauða og agnaáti. Áhugavert verður að greina í framhaldinu hvert hlutverk ATG7(2) er og hvort áður greind hlutverk próteinsins sem eru óháð sjálfsáti gætu verið í gegnum virkni ATG7(2). Greining á ATG7(2) gerir einnig mögulegt að rannsaka ATG7-óháð sjálfsát, sem hefur verið lýst en er lítið þekkt. ATG7 er mikilvægt sjálfsátprótein sem er nauðsynlegt spendýrum og veitir greining á mismunandi ísóformum próteinsins okkur nauðsynlegar upplýsingar um sjálfsátsferlið og tengsl þess við aðra frumuferla.
