    Margar rannsóknir hafa verið gerðar með slímhimnu viðloðandi fjölliðuhimnur til lyfjagjafar í munnholi. Munnholið er þakið slími sem gerir það að verkum að slímhimnuviðloðandi fjölliður þykja ákjósanlegar til notkunar staðbundið í munnholi. Með aukinni slímhimnuviðloðun lyfjaforms er hægt að lengja viðverutíma lyfs í munnholi og ná þannig að auka virkni þess. Doxýcýklín er lyf sem hefur verið rannsakað mikið í þessu samhengi. Rannsóknir hafa sýnt að doxýcýklín hefur hamlandi áhrif á metallópróteinasa (MMP), ensím sem greinast í munnangri, og hefur verið sýnt fram á að meðferð með doxýcýklíni flýtir fyrir bata. Doxýcýklín er viðkvæm sameind og þá sérstaklega í snertingu við vatn, því væri ákjósanlegt að hanna lyfjaform þar sem doxýcýklín væri í vatnsfríu umhverfi.
    Í þessari rannsókn var skoðað hvaða áhrif það hefði á eiginleika fjölliðuhimna með doxýcýklín að framleiða fjölliðuhimnur með vatnsfríum leysi. Kannað var hvaða áhrif það hefði að komplexa doxýcýklín með etýlendíamíntetraediksýru (EDTA) og hýdroxýprópýl-β-sýklódextríni (HP-β-CD). Einnig var doxýcýklín míkróhúðað með karboxýmetýlsellulósu (CMC) með úðaþurrkun og því komið fyrir í fjölliðuhimnum. Við úðaþurrkun fást betri heimtur með hækkuðu hitastigi og virtist úðaþurrkun við hátt hitastig ekki hafa neikvæð áhrif á doxýcýklín.
    Ýmsar prófanir voru gerðar á þeim fjölliðuhimnum sem framleiddar voru. Leysnihraði doxýcýklíns úr himnum var skoðaður og borið saman himnur með mismunandi mýkingaefnum. Viðloðunarkraftur himna var skoðaður og gat verið mikill munur milli himnugerða eftir því hvaða mýkingaefni voru í himnum. Bólgnun himna var skoðuð og kom í ljós að himnur með vatnsfríum leysi hafa góðan bólgnunarstuðul þó þær sundrist hægt, meðan að himnur með vatn sem leysi sundruðust fljótt. Við geymsluþolsmælingar komu upp vandamál og er ljóst að gera þarf frekari rannsóknir til að kanna stöðugleika doxýcýklíns í fjölliðuhimnum.

  • Many studies have been conducted with mucoadhesive polymer films intended for administration of drugs in the oral cavity. The oral cavity is coated with mucus which renders mucoadhesive polymers ideal for topical use in the oral cavity. With increased mucoadhesiveness of pharmaceutical formulations it is possible to extend the residence time of a drug in the oral cavity and thereby increasing its activity. Doxycycline is a drug that has been studied in this context. Studies have shown that Doxycyline has an inhibiting effect on metalloproteinases (MMPs), enzymes detected in mouth ulcers, and it has been shown that administration with Doxycycline speeds up recovery. Doxycycline is a vulnerable molecule and especially in contact with water, therefore it would be desireable to design formulations where Doxycycline was in an anhydrous environment.
    This study explored what impact it would have on polymer films with Doxycycline to produce them with an anhydrous solvent. It was investigated what impact it would have to form complexes of ethylenediamine tetraacetic acid (EDTA) and hydroxypropyl-β-cyclodextrin (HP-β-CD) with Doxycycline. Doxycyline was also microencapsulated using a spray dryer with carboxymethylcellulose (CMC) and the product placed in a polymer film. Spray drying at high temperatures did not seem to have a negative effect on Doxycycline.
    Various tests were performed on the polymer films. Dissolution profile was examined and results compared for films with different plasticizers. The mucoadhesive strength was inspected and it was observed that there could be a significant difference based on what plasticizer was used. Swelling studies of films showed that membranes with an anhydrous solvent had a good swelling index even though they did not disintegrate too easily, while films with water for solvent disintegrated fast. Problems arose while conducting stability testing, further research is needed to examine the stability of Doxycycline.
