    Í stofnasafni Auðlindardeildar Háskólans á Akureyri er að finna yfir 274 bakteríustofna af Pseudomonas ættkvíslinni. Pseudomonas ættkvíslin inniheldur bakteríur sem eru á meðal þeirra hæfustu til að taka sér bólfestu í nálægð við plöntur hvort sem það er í rótarhvolfinu eða á yfirborði þeirra. Ákveðnar bakteríutegundir innan ættkvíslarinnar eru þekktar fyrir að vera sýkjandi fyrir plöntur og þekktasti plöntusýkillinn sem hefur hvað mest látið á sér bera er Pseudomonas syringae.
    Fyrri rannsóknir sýndu fram á að hluti Pseudomonas bakteríustofna innan stofnasafnsins inniheldur sóttburðargen sem tjá fyrir seytikerfi af gerð III (e. type III secretion system) og þá liggur einnig grunur um að á meðal stofnanna sé að finna tegundina P. syringae. Í þessu verkefni er leitast við að fá frekari vísbendingar um plöntusýkingarhæfni Pseudomonas bakteríustofna innan stofnasafnsins. Ákveðnir stofnar eru skoðaðir í nærveru plöntuefna og lífeðilsfræðilegir eiginleikar kannaðir í framhaldinu af því. Áframhaldandi PCR-skimun er gerð eftir sóttburðargenum sem tilheyra seytikerfi af gerð III til frekari staðfestingar á plöntusýkingarhæfni þeirra. Að lokum er gerð PCR-skimun með vísum sem eru sértækir fyrir tegundina P. syringae.
    Niðurstöður verkefnisins leiddu í ljós að nærvera plöntuefna sýndi fram á hvetjandi áhrif á örveruþekjumyndun ákveðinna Pseudomonas bakteríustofna og ákveðnir stofnar framleiddu aðeins yfirborðsvirk efni í nærveru plöntuefna. Af 16 bakteríustofnum bera 7 sóttburðargen sem tjá fyrir mikilvægum þáttum í seytikerfi af gerð III sem staðfestir plöntusýkingarhæfni þeirra. Skimun með Psy-PCR greindi 5 af 16 bakteríustofnum sem skimaðir voru sem P. syringae.

  • The Microbial Culture Collection at the University of Akureyri includes over 274 bacterial strains of the Pseudomonas genus. The Pseudomonas genus has species that are found to be among the most competent bacteria to colonize plants and their immediate vicinity, such as the rhizosphere or phyllosphere. Certain species within the genus are known to be infectious to plants and the most economically important best-studied bacterial plant pathogen is Pseudomonas syringae.
    Previous studies demonstrated that a few of the Pseudomonas strains from the Microbial Culture Collection have virulence genes that encode the type III secretion system. There is also a suspicion that the species Pseudomonas syringae can be found among the Pseudomonas strains in the culture collection. This project seeks for more indications for plant pathogenicity of the Pseudomonas strains. Certain bacterial strains are examined in the presence of plant extracts and specific physiological features are determined. PCR screening for virulence genes of the type III secretion system is performed for further confirmation of plant pathogenicity and another PCR screening with specific primers for identification of P. syringae.
    The results of this project revealed that the presence of plant extracts enhances a biofilm formation of certain Pseudomonas strains and furthermore certain strains only produced biosurfactants in the presence of plant extracts. Seven of the 16 bacterial strains tested had the virulence gene that encodes important components of the type III secretion system, strongly suggesting their plant pathogenicity. Psy-PCR screening identified five of the 16 bacterial strains screened as P. syringae.
