  • Flúrljómunarsmásjár eru meðal mikilvægustu rannsóknatækja í vefjafræði, frumulíffræði, sameindalíffræði og skyldum greinum. Sífellt eru að koma á svið ný tæki og nýjar aðferðir sem leitast við að auka þekkingu manna á grunnþáttum innan líffræðinnar og svo lengi sem til eru spurningar verður þörfin á svörum til staðar. Við Raunvísindastonfun Háskólans hefur verið þróuð ný fljúrljómunartækni sem nota má til háskerpu greiningar á frumum. Tæknin byggir á notkun örflögu sem býr yfir þeim eiginleika að ljósörva einungis þunnt lag nálægt yfirborði hennar (100-2000 nm). Frumum, með flúrljómandi sameindum, er komið fyrir á örflögunni og ljósörvun á sér stað eingöngu í þeim hluta frumunnar sem næst er yfirborðinu. Með þessum hætti má bæði auka greinigæði mynda og skoða einstakar fljúrljómandi agnir innan frumunnar sem veita m.a. upplýsingar um efnaflutning í eða við frumuhimnu. Með örflögunum má ná fram jafnri lýsingu yfir stór svæði, notast má við margar bylgjulengdir til örvunar og hægt er að hanna flögurnar með mismunandi örvunardýpt sem nær frá um 100 nm til 2000 nm. Engin sambærileg tækni er til staðar á Íslandi í dag, en í samstarfi við Lífvísindasetur Læknagarðs (hwww.lifvisindi.hi.is) er vonast til að tæknin bjóði upp á nýja möguleika í rannsóknum á frumum sem leiði til nýrra og mikilvægra upplýsinga um hegðun þeirra.

    Optical devices based on evanescent-wave excitation or sensing are attaining more and more attention in various fields of research and industry. For sensing, the evanescent wave of such devices is exploited to detect changes in the refractive index of the medium into which the wave penetrates. When used for excitation, the short penetration depth of the evanescent field is used to confine the excitation to very small volumes and increasing the signal to background ratio. Numerous practical devices utilize the evancescent-wave principle in one way or another to realize e.g. high sensitivity biosensors or for fluorescence excitation. With the ever growing demand for inexpensive, compact, low power and highly sensitive devices, an increasing emphasis is being put into research and development in this field.
    In this thesis, the design, fabrication, development and characterization process of a novel symmetrical optical polymer based waveguide structure is described. The structure is devised to be used in an aqueous environment and can either be used for evanescent-wave sensing or for excitation. Here, the main focus is put on the excitation capabilities of the device, especially with respect to utilization in fluorescence microscopy. The novelty of the device presented here lies in the symmetric configuration of the structure which gives it some interesting properties. These include the possibility of tuning the penetration depth by varying the structure geometry, efficient end-fire excitation from optical fibers or on-chip light sources and multiple-wavelength excitation through the same optical path. Furthermore, it can provide continuous and uniform surface-confined illumination of macroscopic areas and it places no special requirements on the detection optics.
    In the thesis, the theoretical outline behind the sensing- and excitation principles of the waveguide structure is presented. The performance of the waveguide structure is compared to standard epi- and confocal fluorescence microscopy techniques and the bio-compatiability and functionality of the device, both with respect to fixed- and livecell imaging, is demonstrated. Experimental characterization and numerical modeling of several types of highly integrated optical components based on the waveguide structure is also established. These include directional couplers, microring resonators and interferometric modulators.
