<dim:field mdschema="dc" element="description" qualifier="abstract" lang="is">Iðrabólga er regnhlífarhugtak yfir bólgusjúkdóma í ristli og smáþörmum, en tvö megin form þess eru Crohn’s sjúkdómur og sáraristilbólga. Núverandi meðferðarmöguleikar eru meðal annars ónæmisstillandi lyf og sterar, en orsök sáraristilbólgu er ekki enn þekkt. Þekkt er að sýklalyfið Azithromycin (AZM), sem er makrólíði, hafi þekjustyrkjandi áhrif sem ekki tengjast sýklalyfjavirkni þess. EpiEndo Pharmaceuticals hafa því þróað afleiður af AZM þar sem sýklalyfjahluti lyfsins hefur verið fjarlægður, og kallast þessar afleiður barríólíðar.&#xd;
Í þessu verkefni voru ristilþekjufrumur notaðar til að meta áhrif barriolíðana á þekjuna í meltingarveginum. Frumurnar voru bæði ræktaðar í “air-liquid” og “liquid-liquid” frumuræktum þar sem styrkleiki þekjunnar var metinn með því að mæla rafviðnám þekjunnar. Til þess að líkja eftir umhverfi sáraristibólgu í frumurækt verða frumurnar meðhöndlaðar með bólguörvandi boðefnum. Viðbrögð frumanna verða skoðuð með því að meta tjáningu ýmissa gena, uppsöfnun fosfólípíða innan frumanna, lífvænleika, frumufjölgun og með mótefnalitun á þéttitengjum.&#xd;
Af þeim 15 lyfjum sem voru prófuð í verkefninu voru 9 sem hvorki höfðu áhrif á frumufjölgun né lífvænleika. Lyfin voru því næst skimuð fyrir þekjustyrkjandi eiginleikum en 6 þeirra hækkuðu rafviðnám frumuþekjunnar ásamt því að ýta undir uppsöfnun fosfólípíða innan frumanna. H&amp;E litun sýndi einnig fram á að þær frumur sem voru meðhöndlaðar með þessum 6 lyfjum urðu stærri samanborið við ómeðhöndlaðar þekjufrumur.&#xd;
Barríólíðar eru nýr flokkur af lyfjum með þekjustyrkjandi eiginleikum. Sex þessarra lyfja sýna efnilegar niðurstöður varðandi þekjustyrkjandi eiginleika þeirra á ristilþekjufrumur í frumurækt. Frekari rannsóknir miða að því að prófa þessi lyf á “organ-on-a-chip” frumuræktunarmódeli og meta þá sameindafræðilega ferla sem liggja á bakvið þekjustyrkjandi eiginleika þessarra lyfja.</dim:field>
<dim:field mdschema="dc" element="description" qualifier="abstract" lang="en">Inflammatory bowel diseases (IBD) are complex inflammatory conditions of the colon and small intestine, with Crohn´s disease and ulcerative colitis being the two main types. Current treatments include immunomodulators and corticosteroids that treat symptoms, but neither the cause nor the compromised intestinal epithelial barrier that plays a critical role in the pathophysiology of ulcerative colitis is known. As the antibiotic macrolide azithromycin (AZM) has been shown to have barrier-enhancing properties separate from its antibiotic properties, EpiEndo Pharmaceuticals has developed non-antibiotic derivatives of AZM, termed BarriolidesTM. In this project, human colon epithelial lines were used to evaluate the effects of the BarriolidesTM on the gut epithelium. Cells were cultured in air-liquid and liquid-liquid cultures, and the epithelial barrier was analysed by measuring transepithelial electrical resistance (TEER). A colitis-like condition was induced by treating cells with a mixture of inflammatory cytokines. Cell responses were examined using gene expression analysis, phospholipid accumulation, viability and proliferation studies, and immunostainings.&#xd;
Of the 15 compounds tested, 9 did not affect cell proliferation and viability. The compounds were then screened for barrier-enhancing properties. Six compounds increased TEER and showed an accumulation of phospholipids in the cells. Hematoxylin and eosin (H&amp;E) staining on cell monolayers treated with the compounds also showed enlarged cells compared to untreated cells. BarriolidesTM are a novel class of drugs with epithelial barrier-enhancing properties. Six show promising effects in enhancing the gut epithelial barrier in culture. Further studies aim to test these compounds on an organ-on-chip assay and evaluate the molecular mechanisms behind the barrier-enhancing effects.</dim:field>
